EDIBLE BERRIES ARE A RICH SOURCE OF ANTHOCYANINS WHICH HAVE BEEN TOUTED FOR THEIR ANTIOXIDANT ACTIVITY AND POTENTIAL TO IMPROVE CARDIOVASCULAR HEALTH. EMERGING RESEARCH FINDS THAT BERRY ANTHOCYANINS HAVE AN ANTI-DIABETIC AND ANTI-OBESITY EFFECT. SEVERAL RODENT STUDIES HIGHLIGHT THE PROTECTIVE EFFECTS OF BERRY CONSUMPTION WHEN PAIRED WITH A HIGH-FAT DIET AND A PLETHORA OF HUMAN CLINICAL WORK HIGHLIGHTS THE INSULIN-SENSITIZING EFFECTS OF DIFFERENT BERRY VARIETIES. WE HAVE DESCRIBED BOTH EFFECTS IN OUR EARLIER CLINICAL WORK; HOWEVER A MAJOR LIMITATION IS THE HIGH VOLUME OF BERRIES REQUIRED TO REACH ANTHOCYANIN DOSES THOUGHT TO ELICIT POSITIVE EFFECTS ON HEALTH. ELDERBERRIES WITH BROAD POTENTIAL FOR CULTIVATION ACROSS THE UNITED STATES ARE A DENSE SOURCE OF ANTHOCYANINS AND OTHER FLAVONOIDS AND ARE A CANDIDATE FOOD TO ADDRESS THE ISSUE OF THRESHOLD ANTHOCYANIN DOSE. OUR PRINCIPAL OBJECTIVE IS TO DETERMINE THE BIOACTIVE POTENTIAL OF ELDERBERRIES IN HUMANS. THE PROPOSED PROJECT AIMS TO DETERMINE THE ABILITY OF ELDERBERRY INGESTION TO AUGMENT OR IMPROVE 3 MEASURES: 1 SUBSTRATE OXIDATION; 2 INSULIN SENSITIVITY; AND 3 THE GUT MICROBIOME IN OBESE STUDY VOLUNTEERS. WE WILL CONDUCT A RANDOMIZED CROSSOVER PLACEBO-CONTROLLED CLINICAL TRIAL WHERE SUBJECTS WILL BE FED APPROXIMATELY A HALF CUP OF ELDERBERRY JUICE OR PLACEBO IN COMBINATION WITH A CONTROLLED HIGH-FAT DIET FOR 7 DAYS. MEASURES ON INDIRECT CALORIMETRY MEAL TOLERANCE AND THE MICROBIOME WILL BE PERFORMED AT THE END OF EACH DIET PERIOD. THIS SEED PROJECT WILL PROVIDE PRELIMINARY DATA ON THE USEFULNESS OF ELDERBERRIES TO ENHANCE DIETARY APPROACHES TOWARDS THE PREVENTION AND/OR IMPROVEMENT OF OBESITY AND INSULIN RESISTANCE. THIS PROJECT ALIGNS WITH AFRI'S PROGRAM AREA TO INVESTIGATE FOOD FOR HUMAN HEALTH.

TETRACYCLINES ARE THE MOST COMMONLY USED ANTIBIOTIC FOR TREATMENT AND PREVENTION OF DISEASE INHUMANS AND LIVESTOCK, INCLUDING HONEY BEES. LIKE OTHER ANTIBIOTICS, TETRACYCLINES HAVE BEEN CLASSIFIED AS A CHEMICAL THAT NEGATIVELY AFFECTS HORMONES BECAUSE OF ITS IMPACT ON REPRODUCTIVE CELLS. IN MAMMALS, OXYTETRACYCLINE HAS BEEN SHOWN TO DECREASE THE MOVEMENT OF SPERM AND THE AMOUNT OF LIVE SPERM, IT CAUSES DAMAGE TO TESTICLES, AND REDUCES THE PRODUCTION OF SPERM AND REPRODUCTIVE FERTILITY OVERALL. HOWEVER, THE IMPACT OF TETRACYCLINES ON HONEY BEE REPRODUCTION HAS NOT BEEN INVESTIGATED. HISTORICALLY, A HONEY BEE QUEEN'S LIFESPAN WAS 2-7 YEARS, BUT NOW 50% OF COLONIES REPLACE THEIR QUEENS WITHIN SIX MONTHS. A COLONY WILL REPLACE THEIR QUEEN FOR SEVERAL REASONS, INCLUDING LACK OF STORED SPERM WITHIN THE QUEEN. COLONIES THAT ARE FAILING HAVE QUEENS WITH LOW SPERM COUNTS AND SPERM COUNTS ARE HIGHLY VARIABLE IN DRONES (THE MALE HONEY BEE), BUT IT IS UNCLEAR WHY. GIVEN THE IMPORTANCE OF QUEEN REPRODUCTIVE SUCCESS TO HONEY BEE COLONY SURVIVAL, IT IS IMPERATIVE TO DETERMINE THE FACTORS CONTRIBUTING TO DIMINISHED REPRODUCTIVE QUALITY IN BOTH QUEENS AND DRONES. ADDITIONALLY, THE GUT MICROBIOME (COMMUNITY OF BACTERIA THAT LIVE IN THE GUT) HAS BEEN SHOWN TO PLAY A ROLE IN GROWTH AND DEVELOPMENT OF HONEY BEE WORKERS BUT ITS IMPORTANCE IN QUEEN AND DRONE HEALTH AND REPRODUCTIVE DEVELOPMENT ARE UNKNOWN. HERE WE AIM TO DETERMINE HOW ANTIBIOTICS IMPACT THE REPRODUCTIVE HEALTH OF HONEY BEE 1) QUEENS, AND 2) DRONES, AND 3) INVESTIGATE THE LINK BETWEEN THE MICROBIOME AND REPRODUCTION IN HONEY BEES.

**AWARDS ISSUED PRIOR TO JANUARY 20, 2025, WERE FUNDED UNDER PREVIOUS ADMINISTRATIONS AND MAY NOT REFLECT THE PRIORITIES AND POLICIES OF THE CURRENT ADMINISTRATION.** DESPITE THE RECOGNITION THAT INSECT MICROBIOMES ARE IMPORTANT IN INSECT HEALTH, FEW STUDIES HAVE DELVED INTO THE IMPORTANCE OF THE INSECT MICROBIOME IN ITS INTERACTIONS WITH PLANTS. IN PREVIOUS WORK, WE DEMONSTRATED THAT THE RUSSIAN WHEAT APHID (RWA) MICROBIOME IS CRITICAL TO APHID VIRULENCE. HOWEVER, WHICH MICROBE AND MICROBE FUNCTIONS WITHIN THE MICROBIOME CONTRIBUTE TO RWA VIRULENCE, AND IF THE MICROBES CONTROL RWA BIOTYPE ARE NOT YET KNOWN. THE WORK PROPOSED HERE WILL DISCOVER BACTERIA IMPORTANT TO THIS INTERACTION AND WILL IDENTIFY THE FUNCTIONAL CAPACITIES OF THE ENTIRE MICROBIOME, AS WELL AS OF THE SPECIFIC MEMBERS OF THE MICROBIAL COMMUNITY THAT ARE IMPORTANT CONTRIBUTORS. A MULTI 'OMICS' APPROACH, INCLUDING AMPLICON SEQUENCING, SHOTGUN METAGENOMICS AND METATRANSCRIPTOMICS, WILL BE USED TO IDENTIFY BACTERIAL SPECIES IN THE APHID MICROBIOME ARE IMPORTANT TO THE SUCCESS OF RWA. THE MULTI 'OMICS' DATASETS WILL BE USED TO LINK MICROBIAL FUNCTIONS TO RWA VIRULENCE AND BIOTYPE USING MACHINE LEARNING AND ROBUST STATISTICAL APPROACHES. THIS INFORMATION WILL BE USED TO CONSTRUCT SYNTHETIC COMMUNITIES (SYNCOMS) WHICH WILL BE ADMINISTERED TO RWA FOR PLANT PHENOTYPING EXPERIMENTS DESIGNED TO ASSESS MICROBIAL CONTRIBUTION TO VIRULENCE AND BIOTYPE. STUDIES UTILIZING MRNA SEQUENCING TO DEDUCE EXPRESSION PROFILES OF NON-OBLIGATORY INSECT-ASSOCIATED BACTERIA ARE IN THEIR INFANCY; THIS IS A FIRST ATTEMPT TO FUNCTIONALLY CHARACTERIZE THE ROLE OF THESE BACTERIAL COMMUNITIES. FURTHERMORE, INFORMATION GENERATED FROM THIS WORK WILL BE THE FOUNDATION FOR HARNESSING THE GENOMIC POTENTIAL OF THE RWA MICROBIOME FOR IMPROVED PEST CONTROL.
