REGISTRATIONS ON SPECIALTY CROPS OR MINOR USES DOES NOT PROVIDE AN ADEQUATE ECONOMIC RETURN TO INDUSTRY.THE NER ACTIVITIES ARE CLOSELY INTEGRATED WITH THE CORRESPONDING ACTIVITIES OF THE OTHER REGIONAL PROGRAMS AND THE USDA-ARS UNITS THAT COMPRISE THE NATIONAL IR-4 PROGRAM. THIS PROJECT SEEKS TO BUILD UPON AND EXPAND CAPACITIES FOR RESEARCH, TEACHING AND EXTENSION BY WIDENING THE INVESTIGATION OF ETHNIC CROP PRODUCTION, IMPROVING PRE- AND POST-HARVEST PEST MANAGEMENT, CONDUCTING PHYTOCHEMICAL AND ANTIOXIDANT ANALYSIS ON THE CROPS, INVESTIGATING VALUE-ADDED PRODUCT DEVELOPMENT TO CREATE NICHE MARKETS, CREATING UNDERGRADUATE COURSES IN FOOD SCIENCE, PROVIDING EXPERIENTIAL LEARNING OPPORTUNITIES FOR STUDENTS IN AGRONOMIC PRODUCTION AND FOOD SCIENCE, AND CONDUCTING TRAINING WORKSHOPS FOR SMALL FARMERS ON SUSTAINABLE PRODUCTION PRACTICES, VALUE-ADDED PRODUCT DEVELOPMENT/PACKAGING/MARKETING, AND POTENTIAL MARKETS. HOWEVER, THE UTILIZATION OF ITS COUNTERPART, EGG YOLK, HAS BEEN CONSIDERABLY RESTRICTED BECAUSE OF ITS HIGH LIPID AND CHOLESTEROL CONTENTS AND LIMITED FUNCTIONALITIES. THE IMBALANCED UTILIZATION OF PROCESSED EGG PRODUCTS BECOMES A THREAT TO THE SUSTAINABILITY OF THE EGG INDUSTRY AND PRICE STABILITY FOR CONSUMERS. THEREFORE, NOVEL WAYS SHOULD BE DEVELOPED TO IMPROVE ITS FUNCTIONAL AND NUTRITIONAL PROPERTIES AND CONSEQUENTLY INCREASE THE UTILIZATION OF EGG YOLK. ONE OF THE PROMISING APPROACHES IS TO EMPLOY FRACTIONATION TECHNOLOGY TO PRODUCE EGG YOLK FRACTIONS WITH UNIQUE FUNCTIONAL AND NUTRITIONAL CHARACTERISTICS. OUR LONG-TERM GOAL IS TO DEVELOP NOVEL FUNCTIONAL AND NUTRITIONAL FOOD INGREDIENTS FROM EGG YOLK TO INCREASE ITS USE AND VALUE. OUR SUMMER REEU TEN WEEKS ANNUAL RESEARCH AND EXTENSION PROJECT INVOLVES NOVEL HORTICULTURAL PHYTOCHEMISTRY STUDIES, USING HIGH-VALUE SPECIALTY CROPS WITH UNIQUE PHYTOCHEMICAL CHARACTERISTICS, CULTURAL MANAGEMENT, PROCESSING, AND NUTRIENT PRESERVATION ASPECTS, SOIL ANALYSIS, AGRICULTURAL STATISTICS, SCIENTIFIC SAMPLING AND OUTREACH TO LOCAL FARMERS. A MAJORITY OF WINE AND TABLE GRAPE CULTIVARS CURRENTLY CULTIVATED WORLDWIDE HAVE BEEN GROWN FOR CENTURIES AND ARE PROPAGATED THROUGH ASEXUAL/CLONAL MEANS USING CUTTINGS, GRAFTING OR BUDDING. ALTHOUGH THESE CULTIVARS POSSESS HIGHLY DESIRABLE VITICULTURAL AND ENOLOGICAL CHARACTERISTICS, THEY ARE EXTREMELY SUSCEPTIBLE TO A NUMBER OF BIOTIC STRESSES INCLUDING FUNGAL, BACTERIAL AND VIRAL DISEASES. THE HETEROZYGOUS NATURE OF THE GRAPEVINE GENOME AND SEVERE INBREEDING DEPRESSION LIMITS TRAIT IMPROVEMENT IN COMMERCIAL GRAPEVINE CULTIVARS WITHOUT CAUSING MAJOR DISRUPTIONS IN EXISTING CHARACTERISTICS. THUS IT IS DIFFICULT TO IMPROVE EXISTING CULTIVARS FOR DESIRED TRAITS SUCH AS DISEASE RESISTANCE. IMPROVEMENTS IN GRAPE PRODUCTIVITY AND QUALITY ARE POSSIBLE WITH A BETTER UNDERSTANDING OF THE MOLECULAR GENETICS OF VINE RESPONSE TO BIOTIC STRESS IN CONCERT WITH GENOME EDITING TECHNOLOGY FOR RAPID DEVELOPMENT OF NEW CULTIVARS. GENOME EDITING TECHNOLOGIES NOW MAKE IT POSSIBLE TO IMPROVE SPECIFIC TRAITS IN GRAPE CULTIVARS WHILE KEEPING EXISTING DESIRABLE CHARACTERISTICS INTACT. WE HYPOTHESIZE THAT DEVELOPMENT OF TRANSIENT GENE EXPRESSION/GENOME EDITING SYSTEMS CAN ENABLE RAPID TRAIT IMPROVEMENT IN GRAPEVINE WITHOUT THE INSERTION AND INTEGRATION OF FOREIGN DNA SEQUENCES IN RESULTING CULTIVARS. BIOINOCULANT PRODUCTS IN AGRICULTURE REPRESENT ONE OF THE FASTEST-GROWING MARKETS IN THE WORLD, WITH AN APPROXIMATE 17% ANNUAL INCREASE AND AN ANTICIPATED MARKET VALUE OF $12 BILLION US BY 2026. NEW REGULATORY CONSTRAINTS PLACED ON THE AGROCHEMICAL INDUSTRY ARE EXPECTED TO REDUCE CONSUMER DEMAND FOR CHEMICAL INPUTS AND POSITION THE MICROBIAL-BASED INOCULANTS INDUSTRY TO ABSORB PART OF THIS CURRENT $250 BILLION MARKET. THE ADOPTION OF BIOLOGICAL TOOLS OFFERS A PROMISING PATHWAY TOWARD MORE RESILIENT, EFFICIENT, AND ECOLOGICALLY RESPONSIBLE AGRICULTURAL SYSTEMS, ULTIMATELY SHAPING THE FUTURE OF SUSTAINABLE FOOD PRODUCTION. HOWEVER, THE VARIABLE EFFICACY OF MICROBIAL INOCULANTS AND OUR LIMITED UNDERSTANDING OF THEIR EFFECTS ON SOIL HEALTH AND THE MICROBIOME HAVE SEVERELY HAMPERED THE WIDESPREAD ADOPTION OF MICROBIAL-BASED PRODUCTS. THIS PROPOSED RESEARCH WILL DIRECTLY CONTRIBUTE TO IMPROVING MICROBIAL INOCULANT SUCCESS AND ADVANCE OUR UNDERSTANDING OF THE INTERANNUAL EFFECTS OF BIOLOGICAL AMENDMENTS IN SOIL. AIMS 1 AND 2 WILL LEVERAGE ECOLOGICAL THEORY TO EXPERIMENTALLY MANIPULATE THE COMPONENTS OF PROPAGULE PRESSURE (E.G., NUMBER OF INOCULATIONS AND NUMBER OF CELLS PER INOCULATION) TO ENHANCE INOCULANT ESTABLISHMENT, IMPROVE PLANT PERFORMANCE, AND UNDERSTAND THE IMPLICATIONS OF MICROBIAL INOCULATIONS ON SOIL HEALTH. THIS STUDY FOCUSES ON TWO DISTINCT ORGANISMAL TYPES (I.E., ARBUSCULAR MYCORRHIZAL FUNGI - RHIZOPHAGUS IRREGULARIS, AND BACTERIA - BACILLUS LICHENIFORMIS) ASSOCIATED WITH MICROBIAL-MEDIATED PHOSPHORUS ACQUISITION IN PLANTS.OUR EXPERIMENTAL APPROACH WILL ASSESS THE EFFECTS OF MICROBIAL AMENDMENTS ON PLANT PERFORMANCE, THE SOIL MICROBIOME, AND A SUITE OF SOIL HEALTH PARAMETERS. WE WILL GROW PLANTS IN A CLIMATE-CONTROLLED GREENHOUSE OVER TWO CONSECUTIVE YEARS. AT THE END OF EACH GROWING SEASON, WE WILL USE HIGH-THROUGHPUT GENOMIC TOOLS (E.G., 16S AND ITS AMPLICON SEQUENCING) TO DESCRIBE THE SOIL MICROBIOME FOLLOWING THE APPLICATION OF BIOLOGICAL AMENDMENTS AT DIFFERENT RATES. ADDITIONALLY, WE WILL EMPLOY A SUITE OF BIOGEOCHEMICAL MEASUREMENTS TO UNDERSTAND THE IMPLICATIONS OF MICROBIAL AMENDMENTS ON SOIL HEALTH AND THE ASSOCIATED TEMPORAL PATTERNS OVER TWO GROWING SEASONS. IN PARTICULAR, THE DATA GENERATED FROM THIS EXPERIMENT WILL DIRECTLY CONTRIBUTE TO IMPROVING MICROBIAL INOCULATION METHODS AND INCREASING MICROBIOME-MEDIATED PHOSPHORUS ACQUISITION IN PLANTS.
