A PRIMARY SOURCE OF CONTAMINATION IN DAIRY PRODUCTS IS THE FORMATION OF BIOFILMS ON THE SURFACES OF MILK MANUFACTURE EQUIPMENT, POSING RISKS TO PUBLIC HEALTH AND ECONOMIC STABILITY. WHILE CHEMICAL DISINFECTANTS ARE WIDELY USED TO TREAT BIOFILMS, THEIR USE CAN LEAD TO SIGNIFICANT ISSUES, INCLUDING THE DEVELOPMENT OF ANTIMICROBIAL-RESISTANT BACTERIA. AN ALTERNATIVE APPROACH INVOLVES USING NATURAL CATIONIC ANTIMICROBIALS, WHICH ARE HIGHLY EFFECTIVE AGAINST BIOFILMS. HOWEVER, THEIR APPLICATION IN DAIRY MANUFACTURING IS CHALLENGED BY THEIR TENDENCY TO INTERACT WITH NEGATIVELY CHARGED RESIDUAL MILK PROTEINS, SIGNIFICANTLY DIMINISHING THEIR ANTIMICROBIAL ACTIVITY. THE OVERALL GOAL OF THIS STUDY IS TO ENABLE THE EFFECTIVE USE OF NATURAL CATIONIC ANTIMICROBIALS FOR STERILIZING DAIRY PROCESSING EQUIPMENT. WE PROPOSE TO DEVELOP A SMART CHARGE-REVERSAL POLYMER (PLYS-DA) THAT CARRIES A NEGATIVE CHARGE UNDER NEUTRAL CONDITIONS TO PREVENT INTERACTION WITH MILK PROTEINS BUT REGAINS A POSITIVE CHARGE IN THE ACIDIC MICROENVIRONMENT OF A BIOFILM. THIS CHARGE- REVERSAL PROPERTY ALLOWS PLYS-DA TO ACT AS AN EFFECTIVE ANTIMICROBIAL AGENT UNDER SUCH CONDITIONS. TO FURTHER ENHANCE ANTIMICROBIAL EFFICACY, PH-SENSITIVE ANTIBIOFILM POLYELECTROLYTE NANOPARTICLES WILL BE FABRICATED THROUGH THE ELECTROSTATIC COMPLEXATION OF PLYS-DA WITH VARIOUS CATIONIC ANTIMICROBIAL POLYPEPTIDES, AIMING TO ACHIEVE A BROADER SPECTRUM OF ANTIMICROBIAL EFFECTS. THE ANTIBIOFILM EFFICACY OF OUR DEVELOPED CHARGE-REVERSAL POLYMER AND PH-SENSITIVE NANOPARTICLES WILL BE THOROUGHLY EVALUATED AND VALIDATED.
