Abstract
This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. In liquid chromatography a number of parameters, such as the stationary phase, mobile phase, mobile phase gradient and column diameter, can affect separation performance. We are interested in investigating the affect of column temperature on peak capacity, peptide separation and peptide recovery. We will design and construct a custom column heater that integrates into our mass spectrometer source stage, and analyze complex protein samples at a variety of temperatures and column resins.
