Abstract
Mutation of genetic DNA is a major factor causing cancer. Locating these changes in the DNA sequence of single molecules is difficult because they cannot be assayed by conventional sequencing techniques. This project will demonstrate sequencing capabilities for single stranded DNA and RNA immobilized on a novel substrate, and demonstrate a novel method for increasing throughput. Sensitivity to mid-sequence lesions and base modifications such as 8-oxoguanosine, base methylation, and other cancer-relevant nucleotide modifications will also be demonstrated.
