** AWARDS ISSUED PRIOR TO JANUARY 20, 2025, WERE FUNDED UNDER PREVIOUS ADMINISTRATIONS AND MAY NOT REFLECT THE PRIORITIES AND POLICIES OF THE CURRENT ADMINISTRATION.** BOVINE MASTITIS AND ENTERITIS ARE TWO OF THE MOST COMMON AND COSTLY DISEASES AFFECTING DAIRY COWS IN THE UNITED STATES. HOWEVER, VERY LITTLE IS KNOWN ABOUT HOW DAIRY COWS ACQUIRE MASTITIS- AND ENTERITIS-CAUSING BACTERIA FROM THE ENVIRONMENT. THE LONG-TERM GOAL OF THIS RESEARCH IS TO DETERMINE THE CAPACITY OF MUSCID FLIES, WHICH PREFERENTIALLY BREED AROUND MANURE OR SOILED BEDDING, TO ACQUIRE AND TRANSMIT DISEASE-CAUSING BACTERIA TO DAIRY COWS. IN OBJECTIVE 1, THE PRESENCE AND ABUNDANCE OF CLINICALLY-RELEVANT BACTERIAL TAXA AND ASSOCIATED VIRULENCE FACTORS WILL BE EXAMINED IN FLY AND MANURE SAMPLES COLLECTED FROM TWO FOCAL DAIRY FACILITIES IN SOUTH CENTRAL WISCONSIN. FLY AND MANURE SAMPLES WILL ALSO BE USED TO GENERATE LONG-TERM COLLECTIONS OF FLY- AND MANURE-DERIVED BACTERIAL STRAINS FOR USE IN GENOMIC STUDIES. IN OBJECTIVE 2, FUNCTIONAL ASSAYS WILL BE USED TO EXAMINE COLONIZATION AND PERSISTENCE OF KNOWN CLINICAL ISOLATES AND FIELD-COLLECTED STRAINS IN THE HOUSE FLY, MUSCA DOMESTICA.RESULTS WILL PROVIDE NOVEL INSIGHTS INTO THE PROCESSES UNDERLYING ENVIRONMENTAL PERSISTENCE AND DISSEMINATION OF OPPORTUNISTIC BACTERIAL PATHOGENS THAT ARE DETRIMENTAL TO COW HEALTH AND PRODUCTION.
