** AWARDS ISSUED PRIOR TO JANUARY 20, 2025, WERE FUNDED UNDER PREVIOUS ADMINISTRATIONS AND MAY NOT REFLECT THE PRIORITIES AND POLICIES OF THE CURRENT ADMINISTRATION.** THE TOBAMOVIRUS TOMATO BROWN RUGOSE FRUIT VIRUS (TOBRFV) IS A GLOBAL PANDEMIC AND SIGNIFICANT AGRICULTURAL THREAT TO THE WORLDWIDE TOMATO INDUSTRY. TOBRFV OVERCOMES ALL KNOWN TOBAMOVIRUS RESISTANCES IN TOMATO, INCLUDING THE DURABLE IMMUNE RECEPTOR TM-22 . PLANT VIRUSES HAVEMOVEMENT PROTEINS (MPS) THAT ARE REQUIRED FOR CELL-TO-CELL MOVEMENT AND SYSTEMIC INFECTION. TO FACILITATE PROPER FUNCTION AND PERPETUATION OF THE VIRAL LIFECYCLE, THE MPS ENGAGE IN COMPLEX PROTEIN-PROTEIN INTERACTIONS WITH HOST PROTEINS. THE PROJECT GOAL IS TO DETERMINE HOW THE INTERACTOME OF THE TOBRFV MP IMPACTS PATHOGENICITY AND EVASION OF IMMUNITY. THE TOBRFV INTERACTOME WAS PROBED, AND INTERESTING CANDIDATE PROTEINS WERE SELECTED FOR FURTHER CHARACTERIZATION. BIOCHEMICAL, CELLULAR, AND GENETIC APPROACHES WILL BE USED TO DETERMINE IF THE CANDIDATE PROTEINS REPRESENT HOST RESISTANCE OR SUSCEPTIBILITY FACTORS (OBJECTIVE 1). WE WILL ASK CANDIDATE-SPECIFIC MECHANISTIC QUESTIONS SURROUNDING MP INTERACTIONS WITH THE HOST INTERACTORS TO FURTHER DETERMINE HOW THEY ARE INVOLVED IN TOBRFV SUSCEPTIBILITY OR RESISTANCE (OBJECTIVE 2). WE WILL MAP REGIONS OF THE CANDIDATE PROTEINS REQUIRED FOR MP INTERACTION TO DETERMINE POTENTIAL TARGETS OF MUTAGENESIS FOR USE IN THE CREATION OF TOBRFV-RESISTANT TOMATOES. UPON PROJECT COMPLETION, WE EXPECT TO GAIN FUNDAMENTAL INSIGHTS SURROUNDING THE TOBRFV MP INTERACTOME UNDERLYING VIRAL PATHOGENESIS AND IMMUNITY. THESE DISCOVERIES COULD LEAD TO THE DEVELOPMENT OF NOVEL TARGETS FOR TOBAMOVIRUS RESISTANCE IN TOMATOES AND OTHER IMPORTANT SOLANACEOUS CROPS.
