MASTITIS IS INFLAMMATION OF THE MAMMARY GLAND THAT CAUSES SEVERE MILK PRODUCTION LOSSES AND ANIMAL WELFARE CONCERNS IN DAIRY COWS. INFACT, MASTITIS IS ESTIMATED TO COSTTHE US DAIRY INDUSTRY 2 BILLION DOLLARS ANNUALLY AND NEGATIVELY IMPACTS MILK SUPPLY TO CONSUMERS.DESPITE SIGNIFICANT ADVANCES IN TREATMENT AND MANAGEMENT STRATEGIES OVER THE LAST SEVERAL DECADES, MASTITIS CONTINUES TO BE A PREDOMINANT CHALLENGE FOR DAIRY PRODUCERS. DYSREGULATED INFLAMMATION AND OXIDATIVE STRESS ARE MAJOR UNDERLYING COMPONENTS OF MASTITIS AND CONTRIBUTE TO THE CHALLENGES OF MANAGING THE DISEASE.THUS, THERE IS A NEED TO DEVELOP NOVEL STRATEGIES TO AMELIORATEDYSREGULATED INFLAMMATION AND OXIDATIVE STRESS AS THESE ARE PRIMARY MECHANISMS OF MASTITIS PATHOPHYSIOLOGY.ISOPROSTANES (ISOP)ARE MOLECULES THAT ARE GENERATED AS A RESULT OFOXIDATIVE STRESS IN DAIRY CATTLE. ALTHOUGH IT IS WELL-ESTABLISHED THATISOP ARE FORMED DURINGOXIDATIVE STRESS, IT IS UNKNOWN IF THEY HAVE ANEFFECT IN COWS DURING THECONDITION. FOR INSTANCE, IT IS POORLY CHARACTERIZED IF ISOP CAN INCREASE OR DECREASE INFLAMMATION DURING TIMES OF OXIDATIVE STRESS. THEREFORE, THIS STUDY AIMS TO DETERMINE IF ISOP CAN ALTER INFLAMMATORY OUTCOMES IN COWS USING A MODEL OF OXIDATIVE STRESS.CELLS WILL BE COLLECTED FROM DAIRY COWS ANDTREATED UNDER CONDITIONS THAT MIMICINFLAMMATION AND OXIDATIVE STRESS DURING MASTITIS. THE CELLS WILL THEN BE TREATED WITH ISOP TO SEE IF THEY ARE ABLE TO MODULATE OUTCOMES. FINALLY, CELLS WILLBE SUPPLEMENTED WITH OMEGA-6 AND OMEGA-3 FATTY ACIDS OR ANTIOXIDANTS TO SEE IF THE EFFECT OF ISOP DURING OXIDATIVE STRESS CAN BE MODULATED. WITH THE RESULTS GENERATED BY THIS STUDY, IT IS EXPECTED THAT WE WILL HAVE A MORE THOROUGH UNDERSTANDING OF HOW INFLAMMATION AND OXIDATIVE STRESS CONTRIBUTE TO MASTITIS. THUS, OUR FINDINGS REGARDING THE MECHANISMS BY WHICH ISOPROSTANES MITIGATE OXIDATIVE STRESS AND INFLAMMATION IN THE MAMMARY GLAND MAY HELP DEVELOP DIAGNOSTIC AND NUTRITIONAL TOOLS TO REDUCE THE IMPACT OF MASTITIS ON DAIRY COW HEALTH AND WELLBEING.
