**AWARDS ISSUED PRIOR TO JANUARY 20, 2025, WERE FUNDED UNDER PREVIOUS ADMINISTRATIONS AND MAY NOT REFLECT THE PRIORITIES AND POLICIES OF THE CURRENT ADMINISTRATION.** FOOD PRODUCING ANIMALS CONTRIBUTE MORE THAN $300 BILLION TO THE US ECONOMY. AMERICA'S FOOD INDUSTRY HAS A $55.5 BILLION FOOD SAFETY PROBLEM. POULTRY PRODUCE IS ASSOCIATED WITH 25% OF ALL THE FOODBORNE DISEASES IN THE US. THE BACTERIA SALMONELLA AND CAMPYLOBACTER ARE THE MAJOR FOOD SAFETY ISSUES IN POULTRY INDUSTRY, ATTRIBUTED TO LACK OF POTENT AND SAFE ORAL VACCINES AND OTHER EFFECTIVE MITIGATION STRATEGIES. NANOTECHNOLOGY BASED VACCINES ADMINISTERED THROUGH DRINKING WATER TO POULTRY IS A PROMISING APPROACH FOR INDUCING LOCAL IMMUNITY IN THE INTESTINES TO REDUCE THESE ENTERIC BACTERIAL PATHOGENS IN PREHARVEST POULTRY. WE HAVE STANDARDIZED PROCEDURE FOR SYNTHESIZING US FOOD AND DRUG ADMINISTRATION APPROVED BIODEGRADABLE AND BIOCOMPATIBLE CHITOSAN POLYMER-BASED NANOPARTICLE DELIVERY OF SALMONELLA VACCINE PROTEINS ORALLY TO BIRDS, WHICH INDUCES SPECIFIC IMMUNITY AND REDUCE THE BACTERIAL LOAD IN THE INTESTINES. IN THIS USDA NIFA FUNDED RESEARCH ACTIVITY OUR GOAL IS TO FURTHER IMPROVE THE PERFORMANCE OF SALMONELLA ORAL VACCINE BY INCLUDING ADDITIONAL IMMUNE POTENTIATORS IN THE FINAL VACCINE FORMULATION. IN ADDITION, APPLY THE SIMILAR VACCINE NANOTECHNOLOGY APPROACH TO DEVELOP THE CAMPYLOBACTER ORAL VACCINE AND EVALUATE IN BIRDS. WE FOCUS OUR ENTIRE STUDY EXCLUSIVELY IN MEAT PURPOSE BROILERS BECAUSE WE HAVE OVER 9 BILLION BROILERS PRESENT AT ANY GIVEN TIME IN THE USA, AND WE DO NOT HAVE ANY EFFECTIVE STRATEGIES IN PRACTICE TO MITIGATE SALMONELLA AND CAMPYLOBACTER IN BROILERS TO ENSURE FOOD SAFETY AND PROMOTE SUSTAINABLE POULTRY PRODUCTION. THE OUTCOME OF THIS PROJECT WILL PROVIDE A NOVEL DRINKING WATER DELIVERABLE SALMONELLA AND CAMPYLOBACTER VACCINE PLATFORM FOR BROILERS, WHICH IS SAFE, EFFECTIVE, AND INEXPENSIVE. THIS STANDARDIZED ORAL DELIVERABLE VACCINE TECHNOLOGY PLATFORM IN FUTURE MAY BE APPLIED SUCCESSFULLY TO OTHER FOODBORNE ENTERIC PATHOGENS OF POULTRY.
