MANAGED HONEY BEE COLONIES ARE SUBJECT TO INCREASINGLY STRESSFUL CONDITIONS, PARTICULARLY IN AGRICULTURAL ENVIRONMENTS, WHICH EXPOSE BEES TO MULTIPLE POTENTIALLY INTERACTING ENVIRONMENTAL STRESSORS; NAMELY PESTICIDES AND REDUCED FORAGE NUTRITION. A MAJOR CONTRIBUTING FACTOR TO THESE LOSSES (~30%) HAS BEEN BLAMED ON POOR QUEEN QUALITY. DESPITE THIS, LITTLE WORK HAS BEEN DONE TO EXPLICITLY UNDERSTAND HOW QUEENS RESPOND TO INTERACTION STRESSORS. THE QUEEN BEE IS THE SOLE REPRODUCTIVE UNIT OF THE COLONY; THEREFORE, HEALTHY AND FECUND QUEENS ARE ESSENTIAL FOR COLONY GROWTH AND SURVIVAL. THE QUEEN IS DIRECTLY CARED FOR BY THE COLONY AND RESEARCH HAS SHOWN THAT QUEEN FECUNDITY CAN BE AFFECTED BY WORKER NUTRITION. PRELIMINARY RESEARCH SUGGESTS THAT PESTICIDES AND FORAGE NUTRITION MAY INTERACT IN SUCH A WAY THAT COLONIES AND THEIR QUEEN MAY NOT BE ABLE TO BENEFIT FROM NUTRITIONAL ENHANCEMENT OF LANDSCAPES. IN A TWO-YEAR RESEARCH EXPERIMENT, I WILL INVESTIGATE HOW COLONY-LEVEL EXPOSURE TO NUTRITIONAL STRESS AND INSECTICIDE EXPOSURE AFFECTS QUEEN EGG PRODUCTION, EMBRYO VIABILITY, AND WORKER QUEEN-CARE INTERACTIONS BY PERFORMING HIGH-RESOLUTION LABORATORY ASSAYS. TO DETECT PHYSIOLOGICAL UNDERPINNINGS OF SHIFTS IN QUEEN FECUNDITY I WILL EXAMINE CASTE SPECIFIC PHYSIOLOGICAL TRAITS AS WELL AS EXPRESSION OF TARGET GENES RELATED TO REPRODUCTION, DETOXIFICATION, AND NUTRITION. THIS RESEARCH WILL PROVIDE KEY INFORMATION ON HOW HIGHLY RELEVANT ENVIRONMENTAL STRESSORS INTERACT TO AFFECT HONEY BEE QUEEN HEALTH; BY FOCUSING ON HOW QUEENS RESPOND TO THESE STRESSES, WE CAN INFORM CONSERVATION EFFORTS FOR EFFECTIVE POLLINATOR HABITAT, IDENTIFY BENEFICIAL MANAGEMENT PRACTICES FOR HONEY BEES, AND INFORM SELECTION REGIMES FOR STRESS-TOLERANT BEE STRAINS.
