OUR OVERALL GOAL IS TO DEVELOP METHODOLOGIES FOR USING MULTIPLE VIRUSES TO EDIT INTACT PLANTS. WE WILL FOCUS ON DEVELOPING A SYSTEM FOR DELIVERING EDITING REAGENTS TEMPLATES INTO LETTUCE USING LETTUCE MOSAIC VIRUS (LMV, A POTYVIRUS), TOBACCO RATTLE VIRUS (TRV, A TOBRAVIRUS), AND BEAN YELLOW DWARF VIRUS (BEYDV, A GEMINIVIRUS). THIS WILL AVOID USING SLOW AND MUTAGENIC TISSUE CULTURE PROCEDURES. LETTUCE WILL BE USED BECAUSE IT IS EXPERIMENTALLY TRACTABLE AND A TOP TEN MOST VALUABLE US CROP. EDITING WILL BE EVALUATED BY CHANGES IN PHENOTYPE AND AMPLICON SEQUENCING. IN THE LONG TERM, WE WILL EXTEND THIS SYSTEM USING THESE VIRUSES TO OTHER CROPS, ORNAMENTAL SPECIES, AND WEEDS AS WELL AS TO OTHER VIRUSES IN EACH OF THESE FAMILIES TO PROVIDE THE TOOLS TO EDIT A BROAD RANGE OF SPECIES.
