THE NUMBER OF EPIDEMICS DUE TO NEWLY EMERGING INFECTIOUS DISEASES OF PRODUCTION ANIMALS HAS INCREASED RAPIDLY OVER THE LAST FEW DECADES. IMPORTANTLY, OVER 85% OF EMERGING INFECTIONS ARE CAUSED BY RNA VIRUSES. THE EFFECTIVENESS OF EPIDEMIC PREPAREDNESS PLANS DEPENDS HEAVILY ON THE RAPID AVAILABILITY OF EFFECTIVE VACCINES TO CURTAIL SPREAD AND DAMAGE. THE EPIDEMIC CAUSED BY THE PORCINE EPIDEMIC DIARRHEA VIRUS (PEDV) IN THE U.S IN 2013 REPRESENTS A TYPICAL EPIDEMIC WHICH RESULTED IN THE LOSS OF OVER A QUARTER OF THE NEONATAL U.S SWINE POPULATION. TO ADDRESS THE NEED FOR TECHNOLOGY FOR THE DEVELOPMENT OF RAPID-RESPONSE VACCINES, THE INFECTIVE PARTS OF PEDV WERE SUBJECTED TO A COMBINATION OF HEAT AND ENZYMATIC TREATMENT, SUCH THAT THE VIRAL STRUCTURE REMAINED INTACT WHILE THE REPLICATIVE ABILITY OF THE VIRUS WAS DIMINISHED. THE DEVELOPED VACCINE COMPLETELY PREVENTED PEDV REPLICATION AND CLINICAL SIGNS IN VACCINATED WEANLING PIGS IN A CHALLENGE MODEL. THE GOALS OF THE CURRENT PROJECT ARE TOTEST THE RAPID-RESPONSE PEDV VACCINE IN SOWS, USING TARGETED AND ENHANCED ORAL DELIVERY METHODS TO INDUCE STRONG IMMUNITY IN SOW'S MILK. WE EXPECT THE SUCKLING PIGLETS BORN TO VACCINATED MOTHERS WILL BE EFFECTIVELY PROTECTED AGAINST CHALLENGE DUE TO PASSIVE TRANSFER OF ANTIBODIES FROM THE SOW VIA COLOSTRUM. THE DEVELOPED METHODS WILL ALSO HAVE BROAD APPLICABILITY TO OTHER RNA VIRUSES.
