CONSUMPTION OF A TOMATO-RICH AND A SOY-RICH DIET HAS BEEN ASSOCIATED WITH DECREASED INFLAMMATION AND REDUCED RISK FOR NUMEROUS INFLAMMATION-RELATED CHRONIC DISEASES. TOMATOES CONTAIN CAROTENOIDS/FLAVONOIDS AND SOY CONTAINS ISOFLAVONES, WHICH MAY BE PARTIALLY RESPONSIBLE FOR THESE BIOLOGICAL EFFECTS. WE HYPOTHESIZE THAT COMPARED TO A CONTROL LOW CAROTENOID VARIETY TOMATO JUICE, OUR NOVEL HIGH LYCOPENE VARIETY TOMATO JUICE WITH ADDED SOY ISOFLAVONES WILL RESULT IN GREATER IMPROVEMENTS IN MARKERS OF INFLAMMATION FOLLOWING LONG-TERM CONSUMPTION IN HUMANS. WE ALSO HYPOTHESIZE THAT METABOLIC SIGNATURES (FROM BOTH TARGETED PHYTOCHEMICALS AND THEIR METABOLITES AS WELL AS UNTARGETED METABOLITE PROFILES) OF THESE INDIVIDUALS WILL REFLECT THE CONSUMPTION OF OUR TOMATO-SOY JUICE. IN A 'CROPS TO THE CLINIC' APPROACH, OUR MULTIDISCIPLINARY TEAM WILL CONDUCT A RANDOMIZED, CROSSOVER STUDY TO COMPARE A 4-WEEK INTERVENTION WITH EITHER THE TOMATO-SOY JUICE OR THE LOW CAROTENOID TOMATO JUICE IN 30 OBESE MALES. CHANGES IN LEVELS OF CAROTENOIDS (PLASMA) AND FLAVONOIDS AND FLAVONOID METABOLITES (URINE) WILL BE STATISTICALLY CORRELATED WITH CHANGES IN BIOMARKERS OF INFLAMMATION. ADDITIONALLY WE WILL USE UNTARGETED METABOLOMICS TO INVESTIGATE HOW CONSUMPTION OF THESE TWO JUICES ALTER GLOBAL METABOLISM IN OUR SUBJECTS. THIS STUDY WILL BE THE FIRST TO EVALUATE THE IMPACT OF TOMATO AND SOY TOGETHER ON INFLAMMATION IN HUMAN SUBJECTS AS WELL AS ON METABOLIC SIGNATURES. IT WILL PROVIDE RATIONALE TO EXPLORE OPPORTUNITIES FOR PRODUCT DEVELOPMENT OF NEW PROCESSED PRODUCTS COMBINING FUNCTIONAL FOODS FOR MAXIMAL BENEFIT.
