Abstract

We developed a method for Single Molecule Tracking in yeast (Ball et al., 2016). Applying Single Molecule Tracking and mRNA smFISH, we established dependence of the search of CUP1 TF Ace1p for specific binding sites from the chromatin remodeler RSC (Mehta et al., 2018). Using stem-loop approach and CUP1 as a model, we collaborated with Heinz Koeppl from Darmstadt University (Germany) to develop a new method of quantification of transcription from live cells based on Bayesian inference. This method provides accurate estimates of the transcription kinetics in individual cells and allows a high-throughput evaluation of the cell population for the systems that are not in steady state and thus they cannot be analyzed by a traditional autocorrelation method (Wildner et al. 2023). Currently we are applying nano-resolution MINFLUX / DNA-PAINT technique to detailed structural analysis and molecular quantification of the CUP1 TS. We plan to correlate molecular concentrations at TS with transcription efficiency studied by stem-loop approach and CUP1 mRNA smFISH.
