Abstract

In the last year the Protein Technology Core has continued to optimize workflows and procedures to accommodate the requirements of increasing collaborations with diverse needs and antigen types. We have successfully generated nano bodies against a variety of antigens for both structural and cell based projects. We have generated nanobodies with nanomolar affinity for their target antigen. These nanobodies have been utilize to extract endogenous protein complexes from human cell lines, interacting partner proteins have been validated by mass spec analysis. Structure solution of protein nanobody complexes has been achieve using both cryoEM and X ray crystallography. We continue to adapt these tools for applications for both basic and clinical research. We have participated in a number of collaborative projects, supplying high affinity protein binders for structural studies, and have and ongoing collaboration to raise high affinity protein binders against a human protein with direct application to disease and treatment. We will continue to adapt our technology and application to the ongoing needs of the CCR, NCI and NIH.
