Abstract

The Flow Cytometry laboratory applied cutting-edge technology to evaluate tumor cell antigen expression in patients undergoing various antigen-directed therapies, including therapeutic antibody and CAR T-cell therapy. The flow cytometric assays are rapid and precise. This improves the ability of NCI investigators to monitor the changes in antigen expression, as well as to investigate the correlation between antibody binding to tumor cells and treatment response. The quantification of antigens during therapy may provide a window to look into pathways disrupted by novel therapeutic agents. Additionally, flow cytometric testing is also utilized to evaluate patients receiving chimeric antigen receptor (CAR) T cell therapy at NCI. It allows for monitoring the effector CAR T cells in the patient, while at the same time monitoring the responses to therapy. Additionally, the Flow Cytometry Unit also continues to support ongoing research of CCR investigators, including the development of new biomarker assay, such as TSLPR, CD123, LANA, CD38, etc.
