Development of CAR T cells targeting T-cell malignancies

Abstract

We have tested a large number of antibodies targeting the gamma-delta TCR, and we have selected the top two binders. Both binders are scFvs derived from monoclonal antibodies. We have extensively tested CARs with different hinge, transmembrane, and costimulatory domains to identify the best CARs for in vivo testing. In vivo testing is now complete, and we have selected an optimal anti-gamma-delta CAR designated MGDL-28Z. An abstract describing this work was presented at the 2024 American Society of Hematology meeting, and a manuscript describing the work has been submitted. We are also developing CARs targeting alpha-beta TCRs to target the alpha-beta TCR-expressing malignancies since these are the most common types of T-cell malignancies. we have constructed a series of CARs and we are making target cell lines for in vitro testing of these CARs.
