Preclinical GEM and GDA Models of Primary and Metastatic Melanoma

Abstract

Progress on the CCR Request for Application (RFA) supported projects that involve studies in CAPR GDA melanoma models included a significant milestone in optimizing a perioperative biologic scaffold-assisted cancer vaccine in a syngeneic melanoma model, in collaboration with CCR's Cancer Innovation Laboratory. CAPR melanoma modelling researchers completed several multi-arm studies aimed at evaluating the impact of Gentrix surgical matrix devices vs. adjuvant potentiation when combined with PD-1 blockade therapies. These data on comparing various biological scaffolds' properties to potentiate anti-tumor response of known checkpoint inhibitors enable follow-up experiments aimed at preclinical analysis in aggressive metastatically recurrent HGF-CDK4 driven allograft melanoma models of anti-PD-1 therapies in combination with tumor vaccines, and/or Gentrix biological meshes. The data obtain in the aforementioned projects are prepared as two collaborative manuscripts. These results demonstrated that surgical mesh inflammation is modulated by the tumor microenvironment and does not impact survival after tumor resection.
