Abstract
To quantify fluxes though different pathways, and measure their relative contribution to formation of D-2HG, we will label specifically glucose, glutamine and other C sources in IDH1 tumors. Comparison with normal cell flux analysis will allow identification of the source of deregulation of intracellular metabolites using mass spec and NMR in vitro while for the in vivo studies we will use hyperpolarized MRI. These studies on mice will set the basis for similar in vivo studies in the clinical settings in which we will track the fate of brain metabolites as a function of disease progression and treatment.
