The effect of therapeutics on mutated IDH1 cell lines and mice tumor models

Abstract

The overall goal of our research plan is to understand the metabolic changes in IDH-mutated glioma as a function of different therapeutics. We want to study this in vitro in established cell lines, as well as other models. The challenge is that IDH-mutated cell lines are hard to find since the mutation is not maintained in cell culture and therefore they fail to represent the true phenotype. The goal of this project is to verify the phenotype of IDH mutated cells existent in the lab in order to establish a model system in which we can test different therapeutics and their mechanism of action. We plan to quantify D-2HG concentration via a colorimetric assay and mass spectrometry. We plan to detect the protein expression levels using Western Blot.
