Abstract

We are focused on novel drivers of the 3q amplicon that play a critical role in promoting tumorigenesis in lung squamous cell carcinoma, head and neck cancer, and ovarian cancer. These novel drivers can serve as targets of therapeutic intervention and we are focused on the mechanisms by which these amplified kinases promote tumorigenesis. We are also focusing on LZK, that is essential for cancer cell viability in HNSCC and LSCC. We have successfully generated numerous novel catalytic inhibitors and PROTACs to target this kinase. We continue to elucidate the mechanisms by which this kinases drives tumorigenesis with an overarching goal of developing LZK inhibitors to treat HNSCC and ESCC cancer patients. Lastly we are developing novel antibody-precision drug conjugates (APDCs) to co-target multiple kinase drivers in head and neck, Breast, Esophageal, and colorectal cancer.
