Abstract
Under mpMRI-ultrasound fusion guidance, we collect biopsies and then use laser capture microdissection to obtain ultra-pure populations of primary tissue for downstream analysis. In patients on active surveillance, we apply exome sequencing, transcriptome profiling and ATAC-seq to uncover molecular distinctions between those whose cancer advances over time and those whose imaging and PSA levels remain stable. For participants in neoadjuvant intense androgen-deprivation trials, we perform whole-genome, whole-exome and whole-transcriptome sequencing at baseline to identify the features that predict which tumors will respond. Finally, in individuals receiving first-line radiation therapy (with or without hormonal support), we combine whole-exome and transcriptome data with Affymetrix microarray results to explore how genomic profiles relate to radiographic response and biochemical-recurrence-free survival.
