Abstract
This phase Ib/II study demonstrates the feasibility and curative potential of simultaneous targeting of multiple survival pathways in diffuse large B-cell lymphoma (DLBCL). DLBCL is a highly heterogeneous disease, which can be subdivided into specific genetic subtypes. Hence, the development of effective targeted therapy is dependent on an understanding of critical survival pathways and the identification of synergistic drug combinations. In this regard, our group has characterized the role of B-cell receptor (BCR) signaling in DLBCL and has identified synergistic drug combinations that target BCR signaling at multiple points, as well as preliminary evidence for the importance of BCL-2 in high-grade B-cell lymphoma (HGBCL). Based on our in vitro work, we have developed and tested a multi-targeted combination regimen (ViPOR), which we show is curative for BCR-dependent non-GCB DLBCL and HGBCL with MYC and BCL2 rearrangements (HGBCL-DH-BCL2).
