Immune suppressor mechanisms in patients with GI cancer

Abstract

We treated patients with cholangiocarcinoma using the combination of a-VEGF/a-CTLA4/a-PD-L1. We obtained PBMC< blood, and tumor biopsies and performed immune analysis using these samples in parallel to murine studies. We observed that treatment led to a BAFF- and interleukin (IL)-12-dependent expansion and rewiring of T regulatory cells (Tregs) toward an anti-tumor T helper-1 (Th-1)-like fragile state. These findings have important implications of our understanding how a-VEGF treatment, which is initially considered to be an anti-angiogenic therapy,has on anti-tumor immune responses.
