Abstract
The goal of this project is to determine the basis for 'MHC-restricted' recognition by T cells. We have now identified that MHC restricted T cell recognition has two molecular bases. First, it is based on LCK sequestration by CD4/CD8 coreceptors. Second, it is based on LAT sequestration by CD4/CD8 coreceptors. In the absence of both CD4 and CD8 coreceptors in CD4-CD8- mice, the thymus selects MHC-independent TCRs. Indeed, we have identified and characterized 4 novel ligands that select such MHC-independent TCRs.
