Carbohydrate Microarray Profiling of Glycan-Binding Antibodies and Lectins

Abstract
We are taking a multi-pronged approach to improve access to anti-glycan antibodies. First, we use our glycan microarray to evaluate the selectivities of existing antibodies to carbohydrates. We recently expressed over 150 antibodies with published sequences and profiled each of them on our array. This study provides a large evaluation of anti-glycan antibody specificity, information that is crucial for selecting the right antibody for a particular application and interpreting results and identifying gaps in the field. Second, we have developed a database of antibodies to carbohydrates (Database of Anti-Glycan Reagents; DAGR) that can be used to find antibodies and information about those antibodies. Recently, we have also developed a database of anti-glycan antibody sequences, which will be added to DAGR soon. Third, we are using our glycan microarray to discover and engineer new antibodies to carbohydrates. We are developing platforms for directed evolution of anti-glycan antibodies and for discovering new antibodies from B cells. Lastly, we have developed two new platforms for profiling glycomes using libraries of DNA-barcoded anti-glycan antibodies.
