Abstract

Our prior analysis of gene expression has led to the identification of several genes upregulated in tumor associated stromal cells, originally called tumor endothelial markers (TEMs). Two of these, TEM5 and TEM8 encode products that are of particular interest because they reside on the cell surface and may therefore be directly accessible to blood-borne therapeutics. In an attempt to understand the functional role of these genes in responses regulated by the tumor-associated stroma, we have generated TEM5 and TEM8 knockout mice. By challenging the gene disrupted mice with tumors and various other pathological insults, we are evaluating the importance of these genes during normal and pathologic physiology. We are also development neutralizing antibody to better understand TEM function in vivo. These studies are helping us to better understand how the tumor microenvironment responds to stress.
