Abstract
A monoclonal antibody to EGFR, has been humanized and designated A10. A10 fragments have been installed into engineered T cells to kill cancer cells that express high levels of EGFR. Various vectors were used to generate transduced T cells. These engineered T cells are cytotoxic for tumor cell lines and PDX samples grown in NSG mice. Full length A10 has been modified with cytotoxic compounds to produce antibody-drug conjugates (ADCs). These ADCs are potently cytotoxic for cell lines with high levels of EGFR expression. Antibodies to NRP1 were generated and several of these are 'blocking' for ligands such as VEGF, Sema3A and PLGF. A possible inhibitory role for these antibodies is currently being studied.
