Abstract

(i) We have shown that the chromosome architectural protein HU determines, in an Growth condition dependent way, the expression of motility genes facilitating swimming and chemotaxis and represses the expression of fimbrae genes that facilitate cell adhesion and biofilm formation. (ii) We have determined the mechanisms of interplay between different transcription factors, GalR, CRP and HU, that regulate expression of the amphibolic gal operon in E. coli to suit the physiological demands of the sugar D-galactose. (iii) By single molecule analysis, we are testing the dynamics of DNA-bound CI regulator of bacteriophage Lambda. that has been proposed to explain pairwise co-operative binding of CI to adjacent operator DNA elements. We are following the dynamisms by double labeling of the CI-dimers by fluorescence and subsequent FRET analysis at the DNA-bound CI.
