Identification of small molecules capable of lowering prion protein expression

Abstract
During this reporting period, the research team has been working to develop a high-throughput screening assay to be done in 384-well plates, we will generate a cell line with HiBiT tagged prion protein to quantify prion protein expression. The HiBiT tag is an 11-amino acid tag that undergoes complementation in the cell with its larger (LgBiT) NanoLuc fragment. Upon complementation of NanoLuc in the cell and addition of the NanoLuc substrate (furimazine), light is generated proportionally to the amount of HiBiT expression, which is tagged to prion. We will use this cell line to screen compound libraries to identify molecules capable of lowering prion protein expression.
