Cytotoxicity profiling of Genesis library in A673 and Jurkat cells

Abstract

During this reporting period, the NCATS team performed confirmatory assays on 479 cherry-picked compounds across a panel of cell lines, including A673 (Ewing sarcoma), Jurkat (T-ALL), and additional control lines. An additional plate was generated based on new analyses of differential cell killing between Jurkat and A673, with a focus on (though not limited to) phenol-containing molecules, as well as the NCI-60 CellMiner FADS2 top-list comparison. These compounds were also tested in the confirmation assay. We further conducted a broader-spectrum activity analysis using SUDHL5 screening data, incorporating additional cancer lines (e.g., HCT116, which harbors common resistance mutations) and normal cell lines to better characterize compound activity profiles. From this analysis, we identified ~40 candidate compounds, which are now prioritized for evaluation on the collaborator’s platform for broader target identification.
