Identification of small molecule inhibitors of USP18

Abstract

During the reporting period, the team developed a high-throughput enzymatic assay to identify small molecule inhibitors of USP18. Screening of several NCATS libraries was implemented and hits were selected for follow-up validation using established activity thresholds cut-offs for further validation in orthogonal and biophysical assays, including the evaluation of compound specificity. The team is also evaluating the implementation of cell-based relevant assays for further characterization of hits.
