Identification of ALPK1 inhibitors

Abstract
During this reporting period we have established high-throughput screening assays to assess activity of ALPK1. We have developed 1) a biochemical assay using truncated recombinant ALPK1 and TIFA substrate; 2) cell activity assays monitoring activation of ALPK1 by ADP-Heptose via a reporter gene; and 3) a HiBiT reporter assay to monitor ALPK1 levels in cells. These assays are being screened against small molecule libraries to evaluate their potential in identifying small molecule inhibitors of ALPK1.
