Abstract

During the reporting period, the project team developed six unique primary assays for the methyltransferases (MTase) NNMT, PNMT, COMT, HNMT, GNMT, and GAMT. ETB scientists have developed a counter-assay using a commercial luminescent methyltransferase kit, and to date have screened over 27,000 compounds from a variety of screening collections to build the activity profiles for each MTase. The project team is also in the process of developing an orthogonal assay using LC-MS to further validate hits of interest. Profiling efforts are expanding to include additional MTase family members, such as TPMT and AS3MT, in the existing profile for this family.
