Abstract
The overarching goal of this program is to identify correlates of protection against H5N1, with an emphasis on the roles of mucosal immune mechanisms in both the upper and lower respiratory tract. Within just six months, the Mucosal Immunology and Virology Unit achieved key milestones: 1. Virus procurement and propagation: We successfully obtained and expanded virus stocks for H1N1, H3N2, H5N1, and RSV, generating titers suitable for in vivo studies. 2. Development of immunological assays: We established and validated key influenza-specific immunological assays, including ELISAs for isotype-specific antibody detection in serum and bronchoalveolar lavage fluid, virus neutralization assays, B and T cell ELIspots, and high dimensional flow cytometry panels to identify and characterize tissue-resident memory T cells and other mucosal immune populations in the respiratory tract. 3. Model establishment: We developed and optimized mouse models for seasonal influenza (H1N1, H3N2), H5N1, and RSV infection, ensuring reproducibility and appropriate pathogenicity for comparative studies. 4. Pilot experiments initiated: Early pilot studies were launched to assess how prior exposure to seasonal influenza viruses impacts the course of subsequent H5N1 infection.
