Abstract

The overarching goal of this project is to develop new tools to combat coronavirus-mediated disease. We have screened plasma from >100 acutely infected and convalescent individuals for targeting of multiple coronaviruses. B cell samples from the top responders were screened for production of antibodies that target different coronaviruses, and we isolated broadly neutralizing antibodies targeting the fusion peptide and stem helix from these individuals. In FY25, we followed up on prior work by identifying next-generation antibodies that are more potent than the initially discovered antibodies, modifying existing antibodies to improve potency against coronaviruses, as well as designing immunogen constructs based on the antibodies that we identified.
