Abstract
In FY25, the Gene Targeting Core successfully provided following services: Scientific consultations to assist with the design and execution of gene-editing projects, as well as cryopreservation, re-derivation and shipment of existing mouse lines were provided to the NIDCR investigators year around. Scientific and technical tranining was provided to a visiting investigator from the University of Chile to provide him with the necessary expertise to open a mouse gene-editing facility at his home institution. Pronuclear injection and/or embryo electroporation service: 11 CRISPR-based gene editing constructs were injected for 7 NIDCR Investigators to generate 8 new geenetically-modified mouse lines. Cryopreservation service: Sperm from 21 mouse models were cryopreserved for 8 NIDCR Investigators and archived in primary and backup storage locations. Rederivation service: 5 sperm straws were thawed and 20 in vitro fertilization experiments were performed to rederive 3 mouse strains for 3 NIDCR Investigators.
