Abstract
We have found that silencing COPA and COPE alters expression of the key heparan sulfate biosynthesis protein NDST1. Using established models of fibroblast senescence and leveraging the expertise in Golgi biology (LNG) and senescent cell proteomics (TGB), we propose to study the effect of silencing COPI genes on Golgi organization, proinflammatory cytokine production, and autophagy. We will then study the mechanisms by which NDST1 and COPI members can regulate cellular uptake and SASP factor production to better understand the impaired transport in the senescent phenotype. Finally, we plan to employ proteomic analysis to identify COPA- and COPE-binding proteins.
