Abstract
In fiscal year 2025, we have continued our investigations of the aging and regenerating liver. We have performed biochemical experiments comparing EZH1 and EZH2 binding and activity assays. We have added a geriatric cohort of mice and then using young, old and geriatric mice compared RNA-seq, ChIP-seq for H3K9me3 and H3K27me3, ATAC-seq and micro-C. Our findings reveal that the geriatric group show strong immune responses with gross reorganizations of euchromatin and heterochromatin. We have also performed single-nuclear multi-omic profiling across five age groups and are currently analyzing the data. Integration of these biochemical and multi-omic data with ongoing functional studies will provide key insights into the molecular mechanisms of liver aging.
