Spatial Multiomic Profiling of the Aging and Regenerating Liver

Abstract

In the past year, we have successfully generated ChIP-seq datasets for H3K4me1, H3K4me2, H3K4me3, and H2BK120ub across 3 timepoints: before liver regeneration and 48 and 72 hours after partial hepatectomy in both young and old mice. Additionally, we have generated bulk RNA datasets spanning 30 mins after surgery to 2 months after surgery. Currently this data is being analyzed to identify specific locations where H3K4me increases in aged livers undergoing regeneration and whether it corresponds to upregulation of specific repressive transcription factors, co-repressors, or repair-related mRNAs. We are also analyzing our spatial multi-ome dataset from a pilot experiment to examine data quality and set up an established pipeline for analysis of additional samples.
