Abstract

In the past year, we have completed spatiotemporal profiling of the aging murine brain across 3 age groups and both sexes using the 10X Genomics Visium platform. Our findings reveal pro-inflammatory signatures near white matter fiber tracts which are particularly strong in females. The transcriptomic signature indicated ongoing events such as microglial activation, astrogliosis, complement activation, and myeloid cell infiltration. Machine learning algorithms identified potential transcription factors driving this pro-inflammatory signature. Our ongoing studies with p300 and CBP suggest that these paralogs are downregulated during aging and in AD mouse models.
