Abstract
IMMUNE MONITORING SHARED RESOURCE: ABSTRACT The study of cellular phenotypes and states within complex biological networks, such as the human immune system, is crucial to the advancement of cancer medicine. Although various tools are available to support this research, significant barriers, such as high costs, complexity in data acquisition, and specialized analytical pipelines often hinder access. The Immune Monitoring Shared Resource (IMSR) was created to provide Mayo Clinic researchers, including Mayo Clinic Comprehensive Cancer Center (MCCCC) members, with comprehensive access to high-dimensional single-cell tools. This initiative aims to accelerate basic, translational, and clinical research by leveraging state-of-the-art technology and advanced expertise to study complex biological systems. These functions are enacted through three Specific Aims: Aim 1: Support high-parameter analysis of single-cell suspensions generated from complex biological systems (high-dimensional cytometry). This service uses cytometry by time-of-flight (CyTOF). Aim 2: Support high-parameter spatial analysis of single-cell data generated from intact tissue sections (high-dimensional spatial biology). Imaging mass cytometry or multiplex immunofluorescence are used to provide this service. Aim 3: Develop, test, validate, and share bioinformatic tools to support analysis of complex datasets generated by using these systems. Close collaboration with the Mayo Clinic Bioinformatics Shared Resource is used to provide this service. The IMSR is an enterprise-wide core that operates under a full-service philosophy, providing MCCCC members with an end-to-end solution that includes project consultations, sample preparation, data acquisition, and data analysis.
