Abstract

Mount Sinai ADRC (Sano): Neuropathology Core (Core D) – Research Summary The Neuropathology Core (NPC) at the Mount Sinai Alzheimer's Disease Research Center (MS-ADRC) is dedicated to advancing Alzheimer's disease (AD) and related dementias (ADRD) research by providing exceptionally well-characterized postmortem human brain tissue and related specimens. The NPC supports both ADRC and non-ADRC researchers, contributing to the understanding of pathological mechanisms and the development of biomarkers and therapies. The objectives are 1. To perform neuropathological evaluations and conduct detailed diagnostic and research assessments using standardized and customized protocols, 2. To maintain an ADRD-focused biorepository by operating a comprehensive biorepository of brain tissues, digital whole slide images, and biospecimens, supporting broad research collaborations, and 3. To provide education and cross-disciplinary support by offering specialized training and research support to educate the next generation of neuropathologists and foster interdisciplinary research. This work is significant because NPC's is essential for developing accurate ADRD diagnoses and understanding disease mechanisms. By supplying high- quality tissues and data, the NPC enables the discovery of biomarkers and therapeutic targets. Educational initiatives address the shortage of specialists and ensure the training of diverse and skilled researchers. This work is innovative because the NPC leverages cutting-edge digital pathology and classical techniques to enhance neuropathological assessments. Innovations include developing new diagnostic criteria, advanced tissue processing protocols, and pioneering digital neuropathology approaches using deep learning and computer vision. Our approach involves close collaborations, providing integrated support for autopsies, data harmonization, biomarker validation, and genetic studies. The NPC's leadership from experienced neuropathologists ensures the Core's effectiveness in supporting diverse research initiatives and fostering community engagement.
