Abstract
PROJECT SUMMARY This proposal seeks funding to acquire four Techniplast Digital VenÆlated Cage (DVC) Isocage NegaÆve systems, a crucial step in upgrading mouse housing and improving husbandry pracÆces while simultaneously enhancing animal welfare in our shared Animal Biosafety Level 3 (ABSL3) Plus Select Agent program at St. Jude Children's Research Hospital (SJCRH). SJCRH's ambiÆous strategic plan for the ï¬scal years 2022â2027 outlines a substanÆal 54% increase in faculty members (from 91 to 140) expected to uÆlize in vivo research resources. This expansion is aligned with the insÆtuÆon's mission to establish a worldâleading research eï¬ort in infecÆous diseases that aï¬ect children. To achieve this goal, we intend to recruit sixteen new faculty members who specialize in laboratoryâbased research on infecÆous diseases and immunity. They will complement the exisÆng faculty of nine, who are engaged in cuÆ«ngâedge research in areas such as inï¬uenza, SARSâCoV2, pneumococcal infecÆons, viral and bacterial therapeuÆcs, and the funcÆons of adapÆve and innate immunity during these infecÆons. AddiÆonally, SJCRH serves as one of the collaboraÆng centers for the World Health OrganizaÆon's Global Inï¬uenza Response and Surveillance System, funcÆoning as the Center for Studies on the Ecology of Inï¬uenza in Animals and Birds. Moreover, our program is funded as a Center of Excellence for Inï¬uenza Research and Response (CEIRR) through grants from the NaÆonal InsÆtute of Allergy and InfecÆous Diseases (NIAID). Considering these commitments and aï¬liaÆons, the ABSL3 facility plays a pivotal role in supporÆng these criÆcal iniÆaÆves. The primary objecÆve of this project is to replace the aging Mouse Biocontainment Individually VenÆlated Caging (IVC) system, which was procured back in 2002. We aim to replace it with the stateâofâtheâart Techniplast DVC system, which oï¬ers realâÆme monitoring of rack blower funcÆons, intraâcage condiÆons, and animal acÆvity. This conÆnuous monitoring will allow for the opÆmizaÆon of husbandry pracÆces and improved veterinary care, ulÆmately leading to more humane endpoints. Furthermore, it will facilitate the Æmely collecÆon of Æssue and ï¬uid samples and data, especially in situaÆons when access to the facility is restrictedâsuch as weekends, holidays, or during severe weatherârelated conÆngencies that can aï¬ect staï¬ng levels. The DVC racks come with four Æmes the backup baÆ©ery life (up to 36 hours) compared to the current system. The DVC system oï¬ers detailed monitoring and immediate noÆï¬caÆon of supply and exhaust air parameters, signiï¬cantly improving our ability to respond eï¬ecÆvely during unforeseen conÆngencies. As a result, this grant proposal promises to substanÆally enhance the operaÆonal eï¬ciency and overall animal welfare in our ABSL3 facility. It will enable us to opÆmize the allocaÆon of resources to meet the increasing demands of current and future invesÆgators at SJCRH.
