Abstract

For this study we developed and optimized a robust, high-throughput (384-well format) ELISA binding assay to quantify Nipah virus (NiV) pre-fusion (pre-F)- and glycoprotein (G)-specific serum antibody binding responses. We also developed and optimized a highly reproducible NiV lentivirus vector-based pseudo-typed neutralization assay for both 96-well and 384-well formats. Serum samples from rhesus macaques immunized with Nipah virus pre-F or G proteins were used to optimize and validate these assays. These NiV ELISA binding and neutralization assays were then used to assess NiV mRNA-1215 vaccine-induced serum antibody responses for binding to pre-F and monomeric G proteins, and for neutralization of NiV pseudo-typed virus in vaccinated African green monkeys (AGM) (study VRC-22-0989) and humans (study VRC322).
