Abstract

COVID 19 related projects: Humanized animals: We are interested in generating humanized NOD/ShiLtJ (hNOD/ShiLtJ) mice for ACE2 gene as a common NIAID resource for underlying conditions in humans for COVID-19 research. We need to change 15 amino acids (AA) in mouse gene with human AA in exon#2 (5 mutations), exon#3 (6 mutations) and exon 9 (five mutations). Using CRISPR/cas gene editing, we have successfully created lines of mice with the mutations in exon#2 and exon# 9. These mice were then used to introduce mutations in the exon#3. We have potentially created new mice with the mutations in exons #2, Exon #3 and exon #9. The DNA from these mice is currently being sequenced. Once confirmed, the animals will be tested for the germline transmission followed by expansion of the colony.
