Abstract
In FY 2023, we investigated the relationship between hot-spots of HPV integration into the human genome in cervical cancer and oropharyngeal cancers and transcriptional hubs in the human genome. Using a combination of chromatin analyses with in silico techniques, we have previously characterized the chromatin and genomic landscape of HPV integration sites in cervical and oropharyngeal cancers. We identified both common fragile sites and super-enhancers as frequent regions of HPV integration. We also identify recurrent integration of HPV genomes at transcriptional regulatory hubs. Ongoing studies are investigating the association of HPV genomes with transcriptional hubs. These studies provide insight into mechanisms of HPV replication, HPV integration and oncogenesis, and help design therapeutic interventions.
