Studying Immune Recognition of Epithelial Cancers Using Organoids

Abstract
We are attempting to grow patient-derived tumor organoid lines (PDTO) and normal tissue organoids from all patients in our adoptive T-cell therapy protocols. We then test their cultured tumor infiltrating lymphocytes or TCR-gene engineered PBL for recognition in vitro of these PDTO. If recognition is not seen in the face of positive recognition controls, we will investigate mechanisms for this immune evasion. We will also investigate whether PDTO can be used for in vitro T-cells sensitization to generate reactivity against common shared tumor neoantigens. Eventually we will also study whether there is a correlation of PDTO recognition and clinical response.
