The role of SMARCA1 in Rhabdomyosarcoma and Skeletal Muscle Differentiation

Abstract

SMARCA1 is expressed highly in RMS tissues but not in muscle tissues. In addition, we show that SMARCA1 interacts with HDAC2 in rhabdomyosarcoma cells. SMARCA1 expression increases during myogenic differentiation. CRISPR/Cas9 was used to knock out SMARCA1 expression in RH30 cells. We are in the process of making SMARCA1 KO RD cells. We will use ATAC-seq to investigate changes in chromatin accessibility as a result of SMARCA1 KO. In addition, we will examine the phenotypic impact of SMARCA1 KO on the differentiation of these PAX-fusion positive (RH30) and negative (RD) cells. The project is in collaboration with Judy Davie.
