Identification of radiosensitizers for prostate cancer

Abstract
The goal of this project is to identify targetable vulnerabilities in prostate cancer that will represent new potential targets or biomarkers that influence current treatments. Using a series of targeted CRISPR/Cas9 libraries, we will target key classes of genes in vivo using a panel of murine syngeneic prostate cancer lines. We are preparing libraries to target hypoxia inducible genes, the DNA damage response, immune modulators and chromatin regulators. Screening will be performed in conjunction with therapeutic treatment of tumors using ionizing radiation, ATR and PARP inhibitors and checkpoint blockade immunotherapy. Identified genes that individually modulate cancer growth or influence treatment outcomes will be prioritized for druggability and further investigated using appropriate approaches.
